Mechanism of Action

Melanotan-II is a synthetic cyclic heptapeptide alpha-MSH analogue with broad melanocortin receptor activity (MC1R-MC5R). Beyond melanogenesis via MC1R, MC3R/MC4R activation in the hypothalamus modulates appetite and sexual arousal. ~1000-fold greater potency than native alpha-MSH.

Key Research Findings

  • Produced significant visible skin darkening at sub-milligram subcutaneous doses
  • Initiated erections in 8 of 10 men with psychogenic ED in double-blind crossover study
  • Served as parent compound from which PT-141 (bremelanotide) was developed
